Plasma choline concentrations were measured in humans, rats, and rabbits within the first few hours of life, before any food was ingested. Neonatal animals and humans had markedly elevated plasma choline levels compared to adult animals or humans. Three repetitive sequence families from the sea urchin genome were studied, each defined by homology with a specific cloned probe one to a few hundred nucleotides long. Recombinant λ-sea urchin DNA libraries were screened with these probes, and individual recombinants were selected that include genomic members of these families. Restriction mapping, gel blot, and kinetic analyses were carried out to determine the organization of each repeat family. Sequence elements belonging to the first of the three repeat families were found to be embedded in longer repeat sequences. These repeat sequences frequently occur in small clusters. Members of the second repeat family are also found in a long repetitive sequence environment, but these repeats usually occur singly in any given region of the DNA. The sequences of the third repeat are only 200 to 300 nucleotides long, and are generally terminated by single copy DNA, though a few examples were found associated with other repeats. These three repeat sequence families constitute sets of homologous sequence elements that relate distant regions of the DNA. 相似文献

A set of four clones containing the two adult β-globin genes of the “single” type mouse C57BL/10 (genotype Hbbs/Hbbs) were isolated from a library of cloned restriction fragments. The two genes, designated βs and βt, were physically mapped onto a 32 kb segment of the chromosome carried by the four clones. βs and βt form a stable heteroduplex 1850 bp long, indicating that they are intact and conserved at this level of resolution throughout their length, including their intervening sequences. The βs gene is allelic with the βdmaj gene of the BALB/ c mouse (genotype Hbbd/Hbbd). These two alleles, as well as their surrounding sequences, are highly conserved. In contrast, heteroduplexes of βt with its BALB/c allele, βdmin, revealed three extensive but localized rearrangements. One region of non-homology falls within the large intervening sequence, IVS2. To the 5′ side of the βt/βdmin gene position two unequal substitutions were observed; each results in the net insertion of about 1000 bp into the Hbbd chromosome. The β/βdmin gene position is bracketed by a 1450 bp inverted repeat. One of the 1000 bp substitutions maps within this inverted repeat. 相似文献

Polyphenoloxidase (PPO; EC 1.14.18.1) activity decreased 8-fold from young to mature Vicia faba L. Moensch (cv. Long pod) leaves. The Km for catechol remained relatively constant from young to mature leaves. Electrophoretic separation and analysis showed that only one active form was present in extracts from various leaf sizes. The amount of this form appeared to decrease with leaf size/age. In extracts which had not tanned, electroblotting and immunostaining indicated that one enzyme form was present with a molecular mass of 45 kDa. Two leaf categories contained greater amounts of this immunological cross-reacting PPO than other leaf categories. When extracts were allowed to darken, immunoblotting detected three enzyme forms with molecular masses of 45, 59 and 63 kDa. The latter two immunological crossracting species had no detectable enzyme activity. Poly-A+ mRNA was isolated from six leaf sizes and translated in vitro. A product corresponding to PPO was present in all leaf categories. Greater amounts of this translation product were observed in medium-sized leaves than in very young or mature leaves. These results suggest that: (1) enzymatic assays for PPO are not reliable indicators of the total amount of PPO protein present in developing leaves, (2) immunoblotting can detect inactive enzyme forms, (3) only one active form of the enzyme is present at all developmental stages, and (4) mRNA corresponding to PPO is present at all developmental stages but appears to be more abundant in certain leaf sizes/ages. 相似文献

Fossil seeds of Ensete, a genus presently native to Asia and Africa, have been recovered from the middle Eocene of Oregon, confirming the presence of Musaceae in the North American Tertiary. The seed of Ensete oregonense sp. nov. is operculate, with a well-defined micropylar collar, a pronounced chalazal chamber, and a wide hilar cavity. A survey of seed morphology in extant Zingiberales provides characters for distinguishing Musaceae from other families of the order, furnishes criteria for distinguishing the three extant genera of Musaceae (Musa, Ensete and Musella), and facilitates critical assessment of fossil seed remains. “Musa” cardiosperma Jain from the Cretaceous/Tertiary boundary Deccan Series of India is excluded from Musaceae (although retained in Zingiberales) on the basis of fruit and seed characters, including the lack of laticifers and absence of a chalazal chamber. We reexamined the musaceous seeds from Colombia that previously were described as Tertiary fossils (Musa enseteformis Berry, 1925) and now believe that they are recent, nonfossil remains, evidently from Ensete ventricosum, which is grown in the region where the specimens were originally obtained. In addition, a reputed fossil banana fruit from the Cretaceous of Colombia was reexamined and determined to be a concretion of nonbiological origin. Ensete oregonense is significant therefore, as the first unequivocal fossil record of Ensete and of Musaceae. Although the Musaceae are currently native only to the Old World tropics, this discovery establishes that the family was present in North America about 43 million years ago. 相似文献

A total of 134 chromosome counts representing 21 taxa of the genus Arnica are presented. Counts are published here for the first time for A. lonchophylla (n = 38) and A. nevadensis (n = 38). Ten previously unreported counts representing 9 taxa are also presented. The basic chromosome number of the genus is x = 19. With respect to chromosome number, the genus exhibits maximum diversity in the Rocky Mountains of Colorado and Wyoming. Arnica angustifolia, A. cordifolia and A. mollis are recognized as mature polyploid complexes, containing several wide ranging polyploid races and only a few highly restricted or scattered diploid races. Within the genus in general, diploids tend to be restricted to unglaciated areas while polyploids are much wider ranging, particularly in unglaciated areas. 相似文献

Comparison of polychlorinated biphenyl (PCB) concentrations between the sexes of mature fish may reveal important behavioral and physiological differences between the sexes. We determined whole-fish PCB concentrations in 23 female summer flounder Paralichthys dentatus and 27 male summer flounder from New Jersey coastal waters. To investigate the potential for differences in diet or habitat utilization between the sexes, carbon and nitrogen stable isotope ratios were also determined. In 5 of the 23 female summer flounder, PCB concentrations in the somatic tissue and ovaries were determined. In addition, we used bioenergetics modeling to assess the contribution of the growth dilution effect to the observed difference in PCB concentrations between the sexes. Whole-fish PCB concentrations for females and males averaged 87 and 124 ng/g, respectively; thus males were 43% higher in PCB concentration compared with females. Carbon and nitrogen stable isotope ratios did not significantly differ between the sexes, suggesting that diet composition and habitat utilization did not vary between the sexes. Based on PCB determinations in the somatic tissue and ovaries, we predicted that PCB concentration of females would increase by 0.6%, on average, immediately after spawning due to release of eggs. Thus, the change in PCB concentration due to release of eggs did not explain the higher PCB concentrations observed in males. Bioenergetics modeling results indicated that the growth dilution effect could account for males being 19% higher in PCB concentration compared with females. Thus, the bulk of the observed difference in PCB concentrations between the sexes was not explained by growth dilution. We concluded that a higher rate of energy expenditure in males, stemming from greater activity and a greater resting metabolic rate, was most likely the primary driver for the observed difference in PCB concentrations between the sexes. 相似文献

Glycosylated α-dystroglycan provides an essential link between extracellular matrix proteins, like laminin, and the cellular cytoskeleton via the dystrophin-glycoprotein complex. In secondary dystroglycanopathy muscular dystrophy, glycosylation abnormalities disrupt a complex O-mannose glycan necessary for muscle structural integrity and signaling. Fktn-deficient dystroglycanopathy mice develop moderate to severe muscular dystrophy with skeletal muscle developmental and/or regeneration defects. To gain insight into the role of glycosylated α-dystroglycan in these processes, we performed muscle fiber typing in young (2, 4 and 8 week old) and regenerated muscle. In mice with Fktn disruption during skeletal muscle specification (Myf5/Fktn KO), newly regenerated fibers (embryonic myosin heavy chain positive) peaked at 4 weeks old, while total regenerated fibers (centrally nucleated) were highest at 8 weeks old in tibialis anterior (TA) and iliopsoas, indicating peak degeneration/regeneration activity around 4 weeks of age. In contrast, mature fiber type specification at 2, 4 and 8 weeks old was relatively unchanged. Fourteen days after necrotic toxin-induced injury, there was a divergence in muscle fiber types between Myf5/Fktn KO (skeletal-muscle specific) and whole animal knockout induced with tamoxifen post-development (Tam/Fktn KO) despite equivalent time after gene deletion. Notably, Tam/Fktn KO retained higher levels of embryonic myosin heavy chain expression after injury, suggesting a delay or abnormality in differentiation programs. In mature fiber type specification post-injury, there were significant interactions between genotype and toxin parameters for type 1, 2a, and 2x fibers, and a difference between Myf5/Fktn and Tam/Fktn study groups in type 2b fibers. These data suggest that functionally glycosylated α-dystroglycan has a unique role in muscle regeneration and may influence fiber type specification post-injury. 相似文献

Prebiotics are non-digestible carbohydrate dietary supplements that selectively stimulate the growth of one or more beneficial bacteria in the gastrointestinal tract of the host. These bacteria can inhibit colonization of pathogenic bacteria by producing antimicrobial substances such as short chain fatty acids (SCFAs) and competing for niches with pathogens within the gut. Pasture flock chickens are generally raised outdoors with fresh grass, sunlight and air, which represents different environmental growth conditions compared to conventionally raised chickens. The purpose of this study was to evaluate the difference in microbial populations from naked neck chicken ceca fed with commercial prebiotics derived from brewer’s yeast cell wall via an Illumina MiSeq platform. A total of 147 day-of-hatch naked neck chickens were distributed into 3 groups consisted of 1) C: control (no prebiotic), 2) T1: Biolex® MB40 with 0.2%, and 3) T2: Leiber® ExCel with 0.2%, consistently supplemented prebiotics during the experimental period. At 8 weeks, a total of 15 birds from each group were randomly selected and ceca removed for DNA extraction. The Illumina Miseq platform based on V4 region of 16S rRNA gene was applied for microbiome analysis. Both treatments exhibited limited impact on the microbial populations at the phylum level, with no significant differences in the OTU number of Bacteroidetes among groups and an increase of Proteobacteria OTUs for the T1 (Biolex® MB40) group. In addition there was a significant increase of genus Faecalibacterium OTU, phylum Firmicutes. According to the development of next generation sequencing (NGS), microbiome analysis based on 16S rRNA gene proved to be informative on the prebiotic impact on poultry gut microbiota in pasture-raised naked neck birds. 相似文献
